A DNA-free subunit herpes simplex virus (HSV) vaccine was administered to 15 volunteers without past evidence of HSV infection and to 25 patients with severe recurrent HSV infection. The immune response to the vaccine in these patients was compared to the immunological status of 20 non-vaccinated control patients with recurrent HSV infection. The vaccine elicited antibody and cell-mediated immunity (CMI) in the 15 subjects without past evidence of HSV infection and this response was similar to that observed after a natural infection. Among the 25 patients who were suffering from recurrent HSV infection the vaccine elicited complement dependent cytotoxic antibodies in 13 of these patients who did not possess these antibodies and increased significantly the titers of these antibodies in the 12 other patients. The vaccine gave a significant increase of the titers of the other specific antibodies as well as the level of cell-mediated immunity. The increase of the immunity level in these latter patient was not due to normal variations since in the non-vaccinated control group the antibody titers and CMI remained stable during the same period of time.

The data on the use of a commercial EIA test system for detection of antibodies in control of preparations against herpes simplex and cytomegaloviruses are presented. The enzyme immunoassay test system for antibody determinations to herpes simplex virus produced by the Odessa bacterial preparations enterprise was shown to be suitable for determination of the specific potency (antigenicity) of herpes simplex vaccine. The advantages of this method over the currently used neutralization test were established. Titration of commercial immunoglobulins detects lots with high litres of antibody to herpes simplex virus. For the same purpose, lots of commercial immunoglobulins were tested for antibodies to cytomegalovirus using a West Germany test-system (Behring). It is concluded that enzyme immunoassay test systems for antibody determinations may be used for screening of lots of immunoglobulins of special effects (against herpes simplex and cytomegalovirus infections) both at the stage of serum and final preparation screening.

Prevention of recurrent genital herpes with the inactivated herpetic divaccine Vitaherpavac against herpes simplex virus types 1 and 2 has a number of advantages over the most commonly used symptomatic therapy: it ceases or significantly reduces the number of recurrences and accordingly prolongs a relapse-free interval, abolishes viremia and the manifestations of clinical symptoms of recurrences, induces no dependence to the vaccine. Coadministration of the Vitaherpavac vaccine and the immunomodulator Giaferon has been shown to have some advantage over vaccination only. The new formulation of the agent as suppositories (per rectum) not only enhances the immunogenicity and protective properties of the vaccine, but also reduces the frequency of its application and makes more convenient for patients to use.
