Antiviral agents. 2. Analogs of 2-(.alpha.-hydroxybenzyl)benzimidazole

Analogs of antiviral 2-(α-hydroxybenzyl)benzimidazole (I) [50-97-5] were prepared from 2-(α-chlorobenzyl)benzimidazole (II) by replacing the active Cl with the appropriate group to yield the desired 2-(α-substituted benzyl)benzimidazole.2-(α-Aminobenzyl)benzimidazole (III) [33100-53-7] at 125 μg/ml caused a 100% decrease of the cytopathic effect of ECHO-12 virus, with a cytotoxic level of 1000 μg/ml for the host cells (a tissue culture of rhesus monkey kidney).2-(α-Aminobenzyl)-1-phenylbenzimidazole (IV) [34919-61-4] gave 100% decrease of the cytopathic effect at 4 μg/ml with the cytotoxic level at 62 μg/ml.

The hydrochlorides of D-(--)(I) and L-(+)-2-(α-hydroxybenzyl)benzimidazole (II) were synthesized by the method of Phillips (CA 23, 141) in which the D-(-) and L-(+) forms of mandelic acid were allowed to condense with o-phenylenediamine in acidic medium.The isomeric benzimidazoles were isolated and purified as their hydrochlorides. I.HCl [m. 210-11° (decomposition), [α]²¹D -82.7° (c 1, H₂O)] was found to be 2.5-3 times more active than II.HCl [m. 210-11° (decomposition), [α]²⁵_D 83.2° (c 1, H₂O)].
