Objective To analyze and identify the chem. components in plasma from Yuxingcao Mixture by UPLC-Q-TOF/MS.Method Acquity BEH C18(100 mm x 2.1 mm, 1.7 μm) was used for isolation and identification of chem. components in Yuxingcao Mixture with a mobile phase of acetonitrile-water containing 0.1% formic acid for gradient elution and the flow rate of 0.2 mL/min.The column temperature was 35 °C, and the sample size was 5 μL.Electrospray ion(ESI) source were applied for the qual. anal. under the pos. and neg. ion mode.The capillary voltage under neg. ion mode was 2 kV, ion source temperature 110 °C, cone hole voltage 30 V, cone hole gas volume flow rate 50 L/h, nitrogen degas temperature 350 °C, degas volume flow rate 800 L/h, and scan range was m/z 50-2000.Result Based on UPLC-Q-TOF/MS anal., a total of 24 compounds(including 15 prototypes and nine metabolites) were identified by comparing with ion chromatograms and pyrolysis information of mass fragmentation of the blank biol. samples and Yuxingcao Mixture samples.Conclusion The prototype components and metabolites identified in the plasma may be the potential active components of Yuxingcao Mixture, which provides a foundation for the establishment of quality markers.
