Analysis of the SlRAF-like B gene family in tomato and the molecular mechanism of SlRAF7 in regulating cold stress resistance

The SlRAF-like B gene family is crucial for the regulation of seed dormancy and response to osmotic stress. In this research, a bioinformatics approach was employed to identify a total of 18 members belonging to the SlRAF-like B gene family within the tomato genome. Phylogenetic analysis has categorized the identified SlRAF-like B genes into four distinct groups, revealing significant differences in conserved motifs and gene structure among the proteins within each cluster. Promoter sequence analysis revealed abundant stress, hormone, and light response elements, suggesting the involvement of SlRAF-like B genes in cold stress responses. RT-qPCR analysis showed that most SlRAF-like B genes are induced by cold stress. A knockout mutant of the SlRAF7 gene, belonging to the SlRAF-like B3 group, was generated and tested under normal and cold stress, demonstrating that SlRAF7 positively regulates cold resistance in tomato plants. Further analysis of antioxidant enzyme activities, expression of related genes, and key cold response genes (ICE1, CBFs, and COR genes) in different genotypes suggests that SlRAF7 may enhance cold resistance by modulating the antioxidant enzyme pathway and the CBF signaling pathway. This study provides initial insights into the physiological and molecular mechanisms that underlie cold stress tolerance in tomato, with a particular focus on the role of the SlRAF7 gene.

ICE1 (Inducer of CBF Expression 1) Is Essential for the Jasmonate‐Regulated Development of Stamen in Arabidopsis thaliana

ABSTRACTFloral organ development, pollen germination and pollen tube growth are crucial for plant sexual reproduction. Phytohormones maintain these processes by regulating the expression and activity of various transcription factors. ICE1, a MYC‐like bHLH transcription factor, has been revealed to be involved in cold acclimatisation of Arabidopsis. This study shows that ICE1 regulates multiple aspects of sexual reproduction, including stamen development, pollen development and germination. Loss‐of‐function mutants of ICE1 exhibit floral organs with shorter filaments, defective anther dehiscence and lower pollen viability compared to the wild type. These abnormalities result in disrupted fertilisation, leading to short siliques, a high rate of seed abortion, and dark, shriveled mature seeds. JAZ proteins (JAZ1 and JAZ9) interact with ICE1, inhibiting its transcriptional activity on jasmonic acid (JA)‐responsive genes, including MYB21, MYB24 and MYB108. This study highlights the essential role of ICE1 as a signalling agent in the JA‐regulated maintenance of sexual reproduction in Arabidopsis thaliana.

Quantifying redox signalling regulatory transcriptional dynamics in Nardostachys jatamansi under abiotic stress response

Understanding the responses of Himalayan medicinal plants to multifactorial stresses is crucial in the face of increasing environmental challenges, primarily characterised by frequent temperature and water availability fluctuations. The present study investigates the physiological, biochemical, and transcript variations in the critically endangered Himalayan medicinal plant Nardostachys jatamansi subjected to cold (15 °C and 10 °C for 30 days), drought (6 % PEG for 30 days), and heat stress (30 °C for 24 h). The primary impact of stress was observed through reduced plant biomass and chlorophyll fluorescence. The effects of abiotic stresses were also evident in the modulation of electrolyte leakage, MDA content and H₂O₂ accumulation. Accumulation of reactive oxygen species was confirmed through DAB and NBT staining, alongside increased DPPH and ABTS radical scavenging activity. Differential expression profiling of the RBOH family transcripts further substantiated the production of ROS. Enhanced enzymatic and non-enzymatic activities were observed under each abiotic stress condition. Additionally, genes specific to the regulatory mevalonate (MVA) pathway (TPS9; HMGR) and the methylerythritol phosphate (MEP) pathway (DXS1; DXR) were found to be differentially regulated. Moreover, differential expression profiling of abiotic stress signalling regulatory transcripts CRLK1, CRLK2, CaM6 and ICE1 was also discovered. These findings provide valuable insights into the physiological and biochemical profiling of N. jatamansi in response to extreme environmental conditions, significantly aiding our understanding of the adaptation strategies of alpine vegetation for their conservation.
