Catalyzes the activation of fatty acids by CoA to produce an acyl-CoA, the first step in fatty acid metabolism (PubMed:10434065). Capable of activating medium-chain fatty acids (e.g. butyric (C4) to decanoic (C10) acids), and certain carboxylate-containing xenobiotics, e.g. benzoate (PubMed:10434065). Also catalyzes the activation of lipoate to lipoyl-nucleoside monophosphate (By similarity). Activates lipoate with GTP at a 1000-fold higher rate than with ATP and activates both (R)- and (S)-lipoate to the respective lipoyl-GMP, with a preference for (R)-lipoate (By similarity).

Label as Equilibrium: A performance booster for Graph Neural Networks on node classification

Graph Neural Network (GNN) is effective in graph mining and has become a dominant solution to the node classification task. Recently, a series of label reuse approaches emerged to boost the node classification performance of GNN. They repeatedly input the predicted node class labels into the underlying GNN to update the predictions. However, there are two issues in label reuse that prevent it from performing better. First, re-inputting predictions that are close to the training labels makes the GNN over-fitting, resulting in generalization loss and performance degradation. Second, the repeated iterations consume unaffordable memory for gradient descent, leading to compromised optimization and suboptimal results. To address these issues, we propose an advanced label reuse approach termed Label as Equilibrium (LaE). It has (1) an improved masking strategy with supervision concealment that resolves prediction over-fitting and (2) an infinite number of iterations which is optimizable within constant memory consumption. Excessive node classification experiments demonstrate the superiority of LaE. It significantly increases the accuracy scores of prevailing GNNs by 2.31% on average and outperforms previous label reuse approaches on eight real-world datasets by 1.60% on average. Considering the wide application of label reuse, many state-of-the-art GNNs can benefit from our techniques. Code to reproduce all our experiments is released at https://github.com/cf020031308/LaE.

Timing of exercise differentially impacts adipose tissue gain in male adolescent rats

OBJECTIVECircadian rhythms of metabolic, hormonal, and behavioral fluctuations and their alterations can impact health. An important gap in knowledge in the field is whether the time of the day of exercise and the age of onset of exercise exert distinct effects at the level of whole-body adipose tissue and body composition. The goal of the present study was to determine how exercise at different times of the day during adolescence impacts the adipose tissue transcriptome and content in a rodent model.METHODSRats were subjected to one of four conditions during their adolescence: early active phase control or exercise (EAC or EAE; ZT13), and late active phase control or exercise (LAC or LAE; ZT23). The effects of exercise timing were assessed at the level of subcutaneous and visceral adipose tissue transcriptome, body composition, hypothalamic expression of orexigenic and anorexigenic genes, blood serum markers and 24-hour core body temperature patterns.RESULTSWe found that late active phase exercise (ZT23) greatly upregulated pathways of lipid synthesis, glycolysis and NADH shuttles in LAE rats, compared to LAC or EAE. Conversely, LAE rats showed notably lower content of adipose tissue. In addition, LAE rats showed signs of impaired FGF21-adiponectin axis compared to other groups.CONCLUSIONSFinally, LAE rats showed higher post-exercise core body temperature compared to other groups. Our results thus indicate that our exercise protocol induced an unusual effect characterized by enhanced lipid synthesis but reduced adipose tissue content in late active phase but not early active phase exercise during adolescence.

Leisure activity engagement attenuates the risks of cognitive impairment induced by unhealthy plant-based diets: a nationwide cohort study

OBJECTIVEWe intended to reveal the joint effects between LAE and uPDI on cognition in Chinese older adults.METHODSData were collected from the Chinese Longitudinal Healthy Longevity Survey. In total, 10,617 individuals aged 65 years and above without cognitive impairment or dementia at baseline were enrolled in 2008 and followed up in 2011, 2014, and 2018. The uPDI and the scores of LAE were derived from survey responses, and both were categorized into three groups (low, intermediate, and high). Individuals with a Mini-Mental State Examination (MMSE) score lower than 18 were considered to have cognitive impairment. Cox proportional hazards models were employed to explore the joint association of uPDI and LAE on cognitive impairment, followed by restricted cubic spline (RCS) to observe the effects of the continuous-type variable of uPDI and the scores of LAE on the risk of cognitive impairment. Stratified analysis was applied to examine the association of LAE with cognitive impairment in uPDI groups (high uPDI vs. low uPDI).RESULTSCompared to participants maintained low scores of LAE and high uPDI, those who maintained high scores of LAE and low uPDI had a decreased risk of cognitive impairment (HR = 0.52, 95% CI, 0.43-0.62). The findings of the stratified analysis demonstrated that the protective effects of high scores of LAE on cognition was pronounced in individuals with low uPDI (HR = 0.61, 95% CI: 0.47-0.79) and those with high uPDI (HR = 0.63, 95% CI: 0.51-0.78).CONCLUSIONSIn this cohort study, a higher score of uPDI, which indicated higher intake of salt-preserved vegetables, sugars, and refined grains, was associated with an increased risk of cognitive impairment, whereas this association may be mitigated by regular engagement in leisure activities.

PNAS丨钟清/张晶/牟姗团队合作揭示RIPK4调控氧化应激坏死和铁死亡新机制
