ATP-dependent transporter of the ATP-binding cassette (ABC) family that actively extrudes physiological compounds, and xenobiotics from cells. Mediates ATP-dependent transport of endogenous metabolites such as cAMP and cGMP, folic acid and N-lactoyl-amino acids (in vitro) (PubMed:10893247, PubMed:12637526, PubMed:12695538, PubMed:15899835, PubMed:17229149, PubMed:25964343). Acts also as a general glutamate conjugate and analog transporter that can limit the brain levels of endogenous metabolites, drugs, and toxins (PubMed:26515061). Confers resistance to the antiviral agent PMEA (PubMed:12695538). Able to transport several anticancer drugs including methotrexate, and nucleotide analogs in vitro, however it does with low affinity, thus the exact role of ABCC5 in mediating resistance still needs to be elucidated (PubMed:10840050, PubMed:12435799, PubMed:12695538, PubMed:15899835). Acts as a heme transporter required for the translocation of cytosolic heme to the secretory pathway (PubMed:24836561). May play a role in energy metabolism by regulating the glucagon-like peptide 1 (GLP-1) secretion from enteroendocrine cells (By similarity).

Protective role of ABCC drug subfamily resistance transporters (ABCC1-7) in intestinal inflammation

The ABCC subfamily contains thirteen members. Nine of these transporters are called multidrug resistance proteins (MRPs). The MRPs have been associated with developing ulcerative colitis (UC). This study aimed to evaluate the ABCC expression in UC patients and its role in a dextran sulfate sodium (DSS)-induced colitis mice model under 5-aminosalicylates or methylprednisolone treatment and compared with control without inflammation. DSS-induced colitis mice were treated with 5-aminosalicylates (50 mg/kg 24 h) or methylprednisolone (2 mg/kg 24 h). Human rectal biopsies were obtained from UC patients. The abcc-relative mRNA levels and protein expression were determined by RT-PCR and immunohistochemistry. abcc4, abcc5, and abcc6 mRNA levels were significantly increased in DSS-induced colitis compared to the other groups. The 5-aminosalicylate treatment dramatically increased the abcc2 and abcc3 mRNA levels vs. control. Methylprednisolone treatment increased abcc1 vs. DSS-induced colitis and colitis treated with 5-aminosalicylate. Immunohistochemical analysis revealed down-regulation of ABCC1/ABCC2/ABCC5/ABCC7 in mice colitis vs. control. Treatment with 5-aminosalicylate restored ABCC5 levels, while methylprednisolone restored ABCC2/ABCC5/ABCC7 in colitis mice at similar control levels. Relative mRNA levels of mrp1-5 were increased in active UC patients vs. control. ABCC2/ABCC4/ABCC7 were conspicuously expressed in the mucosa of 5-aminosalicylate and/or methylprednisolone-treated UC patients, while ABCC2/ABCC4/ABCC5/ABCC7 in submucosa, ABCC1/ABCC5/ABCC7 in muscular, and ABCC1/ABCC4/ABCC5/ABCC7 in serosa were expressed vs. controls. This is the first report about the differential up-regulation of the ABCC subfamily gene and protein expression in DSS-induced colitis under aminosalicylates or methylprednisolone treatment.

Transport of statins by multidrug resistance-associated proteins 1 and 5

Statins are widely used in the treatment of hypercholesterolemia but also associated with muscle-related adverse effects. Multidrug resistance-associated protein (MRP) 1 and 5 are expressed in the skeletal muscle, where they may regulate intramuscular levels of their substrates. Here, we investigated the transport of various statins by MRP1 and MRP5 with the vesicular transport assay. Statin concentrations in the vesicles were determined with liquid chromatography tandem mass spectrometry. At 6 µM statin concentration, MRP1 transported both 3R,5S-fluvastatin and 3S,5R-fluvastatin with uptake ratios of 2.6 and 2.0. MRP5 transported 3R,5S-fluvastatin, 3S,5R-fluvastatin, and 10 µM pitavastatin with uptake ratios of 2.9, 3.7, and 2.6, respectively. Atorvastatin was only a weak substrate of MRP5 with an uptake ratio of 1.6 and was therefore not investigated further. In concentration-dependent transport experiments, racemic fluvastatin was transported by MRP1 and MRP5 with apparent affinities (K_m) of 225 µM and 23 µM. Pitavastatin was transported by MRP5 with a K_m value of 433 µM. In vitro clearance (CL_{in vitro}) of fluvastatin was 0.36 µl/min/mg for MRP1, while MRP5 exhibited a CL_{in vitro} value of 1.2 µl/min/mg for fluvastatin and 0.21 µl/min/mg for pitavastatin. Pravastatin, rosuvastatin, and simvastatin acid were not transported by MRP1 or MRP5. Atorvastatin and pitavastatin were not transported by MRP1. These data indicate that MRP1 transports fluvastatin, while MRP5 transports both fluvastatin and pitavastatin. Because MRP1 and MRP5 are expressed in the skeletal muscle, they may reduce myocyte exposure to fluvastatin and pitavastatin and protect from muscle toxicity.

Exploring the Molecular Intersection of Posterior Ocular Tuberculosis: Mycobacterium tuberculosis Proteins, Ocular Autoimmunity, and Immune Receptor Interactions

PurposeThe presentation of posterior ocular tuberculosis (TB) varies greatly along with the need for immunomodulatory therapy to control inflammation. In this study, we explore the potential mechanisms and pathways for autoimmune-related inflammation in ocular TB using molecular mimicry-based mathematical modeling.DesignComputational protein analysis.MethodsTwenty-three TB-related proteins, including ESAT-6 subgroup proteins, and 23 retinal ganglion cells, photoreceptor, and retinal pigment epithelium (RPE) cellular proteins were included in this study. The 3-dimensional structure and sequence of the TB AG proteins were compared to the above-mentioned retinal, photoreceptor, and RPE cellular proteins. All retinal proteins were obtained from the UniProt database. The sequence and 3-dimensional structure of TB-related proteins and retinal proteins were compared with the TM-align server. The interactions of proteins showing significant similarity (template modeling score above 0.5, root mean square deviation [RMSD] value below 5A°) with cytokines (interleukin [IL]6, IL10, IL12A, IL12B, TLR2, TLR3, and TLR4) were analyzed. Autoimmune and autoinflammation-related protein-receptor interaction of similar proteins was assessed using the CABS-dock web server.Main Outcome MeasuresTemplate modeling score, structural alignment accuracy using RMSD value, protein-cytokine interaction.ResultsWe detected a high level of structural similarity between ESAT-6 (EsxA, EsxB) proteins and rhodopsin, HSPA1A, RPE-related BEST-1, ABCC-1, ABCC-4, ABCC-5, SLC47A1, SLC1A5, SLC38A7, SLC6A6, SLC5A6, LAT-1, and SLC16A1 proteins. When we evaluated the likelihood/potential to stimulate an immune response via a cytokine release, TLR-2 (most common), TLR-3, and TLR-4, which are highly susceptible to Mycobacterium tuberculosis ESAT-6 (ESXA and ESXB) proteins, showed a potential receptor-protein interaction with retinal proteins. Moreover, some eye-related proteins had the capacity to trigger the T-cell response by binding to cytokines such as IL-12, IL-10, and IL-6, which are all highly overexpressed in TB infections.ConclusionsOur study demonstrates that TB proteins may have significant structural similarities with many eye-related proteins. MSLN：超越实体瘤的免疫治疗靶点
