Functions as a ubiquitin E3 ligase and as an ISG15 E3 ligase (PubMed:16352599). Involved in innate immune defense against viruses by mediating ubiquitination of RIGI and IFIH1 (PubMed:17392790, PubMed:29357390, PubMed:30193849, PubMed:31710640, PubMed:33849980, PubMed:36045682). Mediates 'Lys-63'-linked polyubiquitination of the RIGI N-terminal CARD-like region and may play a role in signal transduction that leads to the production of interferons in response to viral infection (PubMed:17392790, PubMed:23950712). Mediates 'Lys-63'-linked polyubiquitination of IFIH1 (PubMed:30193849). Promotes ISGylation of 14-3-3 sigma (SFN), an adapter protein implicated in the regulation of a large spectrum signaling pathway (PubMed:16352599, PubMed:17069755). Mediates estrogen action in various target organs (PubMed:22452784). Mediates the ubiquitination and subsequent proteasomal degradation of ZFHX3 (PubMed:22452784). Plays a role in promoting the restart of stalled replication forks via interaction with the KHDC3L-OOEP scaffold and subsequent ubiquitination of BLM, resulting in the recruitment and retainment of BLM at DNA replication forks (By similarity). Plays an essential role in the antiviral activity of ZAP/ZC3HAV1; an antiviral protein which inhibits the replication of certain viruses. Mechanistically, mediates 'Lys-63'-linked polyubiquitination of ZAP/ZC3HAV1 that is required for its optimal binding to target mRNA (PubMed:28060952, PubMed:28202764). Mediates also the ubiquitination of various substrates implicated in stress granule formation, nonsense-mediated mRNA decay, nucleoside synthesis and mRNA translation and stability (PubMed:36067236).

TRIM25 facilitates ferroptosis in ovarian cancer through promoting PIEZO1 K63-linked ubiquitination and degradation

BACKGROUNDOvarian cancer represents a significant threat to women's health. and ferroptosis is recognized as a potential natural inhibitor in cancer therapy, the regulatory mechanism of TRIM25 in ovarian cancer and its potential for regulating ferroptosis as a treatment remain unclear.METHODSThe role of TRIM25 in ovarian cancer was examined through functional gain- and loss-of-function assays both in vitro and in vivo, while its target genes were identified. The stability and ubiquitination sites of PIEZO1 were analyzed using protein docking and ubiquitination experiments.RESULTSTRIM25 is highly expressed in ovarian cancer and promotes the growth and metastasis of ovarian cancer cells both in vivo and in vitro. Mechanistically, it facilitates PIEZO1 degradation through ubiquitination-dependent proteasome activity, inhibits ferroptosis, and stimulates ovarian cancer cell growth.CONCLUSIONOur study clearly shows that TRIM25 stimulates ovarian cancer by inducing K63-linked ubiquitination of PIEZO1, which suppresses ferroptosis and promotes excessive proliferation of ovarian cancer cells. Further research identified the ubiquitination modification site on PIEZO1, providing insights for ovarian cancer treatment.

Exogenous RNA surveillance by proton-sensing TRIM25

Exogenous messenger RNAs (mRNAs) require cellular machinery for delivery and translation but also encounter inhibitory factors. To investigate their regulation, we performed genome-wide CRISPR screens with in vitro–transcribed mRNAs in lipid nanoparticles (LNPs). Heparan sulfate proteoglycans (HSPGs) and vacuolar adenosine triphosphatase (V-ATPase) were identified as mediators of LNP uptake and endosomal escape, respectively. TRIM25—an RNA binding E3 ubiquitin ligase—emerged as a key suppressor inducing turnover of both linear and circular mRNAs. The endoribonucleases N4BP1 and KHNYN, along with the antiviral protein ZAP, act redundantly in TRIM25-dependent surveillance. TRIM25 specifically targets mRNAs delivered by endosomes, and its RNA affinity increases at acidic pH, suggesting activation by protons released from ruptured endosomes. N1 -methylpseudouridine modification reduces TRIM25’s RNA binding, helping RNAs evade its suppressive effect. This study comprehensively maps cellular pathways regulating LNP-mRNAs, offering insights into RNA immunity and therapeutics.

Dietary Zinc activates the Nrf2 signaling pathway to inhibit pyroptosis and attenuate the lung inflammatory response in COPD

Pyroptosis and inflammation play crucial roles in the development of chronic obstructive pulmonary disease (COPD), and Zinc deficiency is commonly observed in COPD patients. In this study, we aimed to explore the impact of Zinc supplementation on pyroptosis and inflammation in a cigarette smoke (CS)-induced COPD mouse model, as well as the underlying mechanisms. The COPD mouse model was established through CS exposure, and mouse pulmonary epithelial cells (MLE-12) were exposed to cigarette smoke extract (CSE) to further validate the effects of Zinc supplementation. CS exposure resulted in significant alveolar wall damage, increased thickening of the alveolar walls, and elevated levels of interleukin-1β (IL-1β), IL-6, IL-18, and tumor necrosis factor-α (TNF-α) in the lung tissues of COPD mice. However, treatment with dexamethasone (a positive control) or Zinc supplementation alleviated these damages. Furthermore, the expressions of pyroptosis markers, including NLRP3, cleaved-Caspase-1, and GSDMD-N proteins, were upregulated in the lung tissues after CS exposure. Zinc supplementation, however, reversed these changes. Additionally, Zinc supplementation upregulated the protein expressions of nuclear factor erythroid 2-related factor 2 (Nrf2), hemeoxygenase-1 (HO-1), and quinone oxidoreductase-1 (NQO-1), and promoted the ubiquitination of Kelch-like ECH-associated protein 1 (Keap1) mediated by tripartite motif 25 (TRIM25) in the lung tissues of CS-induced mice. Importantly, the Nrf2 signaling inhibitor ML385 abolished the beneficial effects of Zinc in CS-exposed mice. Similar results were observed in MLE-12 lung epithelial cells exposed to CSE. In summary, Zinc supplementation inhibits pyroptosis and attenuates inflammation in COPD mice by activating the Nrf2 pathway.Supplementary InformationThe online version contains supplementary material available at 10.1007/s10616-025-00725-7.

Cell Death & Disease | 复旦大学李继喜团队揭示细胞死亡关键蛋白ZBP1在抗病毒免疫中的作用机制

2024年7月，复旦大学李继喜教授团队在Cell Death & Disease杂志发表了题为“ZBP1 condensate formation synergizes Z-NAs recognition and signal transduction”的研究论文。该研究通过体外和细胞实验证明，ZBP1的Zαβ结构域能结合不同长度的d(CG)n配体；HSV或IAV病毒感染后，Zαβ能够形成具有液液相分离的凝聚体。ZBP1的RHIM结构域形成典型淀粉样凝聚体，其核心基序206IQIG209突变会抑制淀粉样纤维的形成，并阻碍细胞程序性坏死发生和抗病毒免疫的应答信号。本研究发现ZBP1不同结构域可形成凝聚体或淀粉样结构，并通过二者之间的协同作用实现Z型核酸（Z-NAs）的识别和下游信号转导，在应对病毒感染的免疫应答过程中发挥关键作用。 Z-DNA结合蛋白1（ZBP1或DAI）是第一个被发现存在于细胞质中的DNA感知蛋白受体。作为固有免疫应答过程中的关键基因，ZBP1通过Zαβ结构域感知Z型核酸刺激，依赖RHIM结构域与RIPK1、RIPK3、TRIF等蛋白结合，参与泛死亡小体（PANoptosome）组装，诱导细胞程序性死亡来抵抗病原体侵染。研究也表明ZBP1与热射病、自身免疫疾病、肠炎、癌症等多种疾病密切相关。本研究表明ZBP1能够通过Zαβ结构域响应体内异常Z-NAs刺激后，形成一个液滴状凝聚体平台，使ZBP1易于通过RHIM结构域进行自身聚集并与下游蛋白（如RIPK1、RIPK3）结合，揭示了ZBP1在应对病原体感染的免疫反应中的具体调控机制。李继喜课题组长期从事细胞死亡与免疫应答研究。已发现坏死小体组分RIP1/RIP3形成功能性淀粉样纤维复合物，作为级联放大平台，激活程序性细胞坏死（2012，Cell）；通过结构生物学方法解析了坏死小体的高分辨率三维结构（2018，Cell）；发现多个程序性细胞死亡的调控分子（TRIM21、TRIM25、SP1），并阐明相关作用机理（2021&2022，Cell Death Differ；2023 Mol Neurodegener; 2024 Cell Death Dis）。图1.
