Peroxisomal and mitochondrial MAVS act sequentially to create an antiviral cellular state (PubMed:20451243). Upon viral infection, peroxisomal MAVS induces the rapid interferon-independent expression of defense factors that provide short-term protection, whereas mitochondrial MAVS activates an interferon-dependent signaling pathway with delayed kinetics, which amplifies and stabilizes the antiviral response (PubMed:20451243). May activate the same pathways following detection of extracellular dsRNA by TLR3 (PubMed:16153868). May protect cells from apoptosis (PubMed:16125763). Involved in NLRP3 inflammasome activation by mediating NLRP3 recruitment to mitochondria (PubMed:23582325).

Transcription of endogenous retroviruses in senescent cells contributes to the accumulation of double-stranded RNAs that trigger an anti-viral response that reinforces senescence.

An important epigenetic switch marks the onset and maintenance of senescence. This allows transcription of the genetic programs that arrest the cell cycle and alter the microenvironment. Transcription of endogenous retroviruses (ERVs) is also a consequence of this epigenetic switch. In this manuscript, we have identified a group of ERVs that are epigenetically silenced in proliferating cells but are upregulated during replicative senescence or during various forms of oncogene-induced senescence, by RAS and Akt, or after HDAC4 depletion. In a HDAC4 model of senescence, removal of the repressive histone mark H3K27me3 is the plausible mechanism that allows the transcription of intergenic ERVs during senescence. We have shown that ERVs contribute to the accumulation of dsRNAs in senescence, which can initiate the antiviral response via the IFIH1-MAVS signaling pathway and thus contribute to the maintenance of senescence. This pathway, and MAVS in particular, plays an active role in shaping the microenvironment and maintaining growth arrest, two essential features of the senescence program.

Potentiating the radiation-induced type I interferon anti-tumoral immune response by ATM inhibition in pancreatic cancer.

Radiotherapy induces a Type I interferon (T1IFN)-mediated anti-tumoral immune response that we hypothesized could be potentiated by a first-in-class ATM inhibitor leading to enhanced innate immune signaling, T1IFN expression, and sensitization to immunotherapy in pancreatic cancer. We evaluated the effects of AZD1390 or a structurally related compound AZD0156 on innate immune signaling and found that both inhibitors enhanced radiation-induced T1IFN expression via the POLIII/RIG-I/MAVS pathway. In immunocompetent syngeneic mouse models of pancreatic cancer, ATM inhibitor enhanced radiation-induced anti-tumoral immune responses and sensitized to anti-PD-L1, producing immunogenic memory and durable tumor control. Therapeutic responses were associated with increased intratumoral CD8+ T cell frequency and effector function. Tumor control was dependent on CD8+ T cells as therapeutic efficacy was blunted in CD8+ T cell-depleted mice. Adaptive immune responses to combination therapy provided systemic control of contralateral tumors outside of the radiation field. Taken together, we show that a clinical candidate ATM inhibitor enhances radiation-induced T1IFN leading to both innate and subsequent adaptive anti-tumoral immune responses and sensitization of otherwise resistant pancreatic cancer to immunotherapy.

Inhibition of Aurora kinase induces endogenous retroelements to induce a type I/III interferon response via RIG-I.

Type I interferon signaling is a crucial component of anti-viral immunity that has been linked to promoting the efficacy of some chemotherapeutic drugs. We developed a reporter system in HCT116 cells that detects activation of the endogenous IFI27 locus, an interferon (IFN) target gene. We screened a library of annotated compounds in these cells and discovered aurora kinase inhibitors (AURKi) as strong hits. Type I IFN signaling was found to be the most enriched gene signature after AURKi treatment in HCT116, and this signature was also strongly enriched in other colorectal cancer (CRC) cell lines. The ability of AURKi to activate IFN in HCT116 was dependent on MAVS and RIG-I, but independent of STING, whose signaling is deficient in these cells. MAVS dependence was recapitulated in other CRC lines with STING pathway deficiency, whereas in cells with intact STING signaling, the STING pathway was required for IFN induction by AURKi. AURKi's were found to induce expression of endogenous retroviruses (ERV's). These ERVs were distinct from those induced by the DNA methyltransferase inhibitors (DNMTi's), which can induce IFN signaling via ERV induction, suggesting a novel mechanism of action. The anti-tumor effect of alisertib in mice was accompanied by an induction of IFN expression in HCT116 or CT26 tumors. CT26 tumor growth inhibition by alisertib was absent in NOD/SCID mice vs. WT mice, and tumors from WT mice with alisertib treatment showed increased in CD8+ T cell infiltration, suggesting that anti-tumor efficacy of AURKi depends, at least in part, on an intact immune response.

Molecular Cell | 生万强/施扬课题组发文揭示MAVS棕榈酰化修饰响应表观干预增强抗肿瘤免疫

表观遗传调控异常促进肿瘤恶性转化和逃避免疫监视。肿瘤细胞中，编码非我抗原或免疫刺激分子的基因常常被表观遗传学机制所沉默，造成“免疫忽视”。广泛分布于细胞基因组中的逆转座子（REs）可以通过内源性双链RNA （dsRNA）的形式产生免疫刺激分子。近年来，包括DNMTs和LSD1在内的多个表观遗传因子被发现可以调控肿瘤细胞中逆转座子来源的dsRNAs的丰度和结构完整性，进而影响肿瘤免疫应答和免疫治疗效果。鉴于这些发现，表观疗法联合免疫疗法的治疗手段目前被应用于临床测试。值得注意的是，靶向上述表观遗传因子主要是通过RIG-I/MDA5-MAVS通路激活I型干扰素应答来发挥免疫激活作用，这提示肿瘤内I型干扰素应答的强度和时程可能对于表观联合免疫疗法的疗效至关重要。相对于目前诸多研究聚焦于发掘更多表观遗传因子调控内源性dsRNA分子的产生，对于细胞质中内源性dsRNA分子的免疫刺激效应是如何被调控的却缺乏研究。2023年12月7日，浙江大学基础医学院/良渚实验室生万强课题组与牛津大学Ludwig癌症研究所施扬教授课题组合作，在Molecular Cell杂志上发表了题为“CPT1A induction following epigenetic perturbation promotes MAVS palmitoylation and activation to potentiate antitumor immunity”的研究论文，从比较表观干预和病毒感染在引起I型干扰素应答的异同入手，细致地揭示了线粒体蛋白CPT1A响应不同刺激信号调控MAVS蛋白S-棕榈酰化修饰的分子机制和产生的重要作用。研究团队发现与病毒感染激活的I型干扰素应答不同，表观干预可以维持MAVS蛋白稳定表达进而持续激活干扰素通路。MAVS是转导dsRNA模式识别受体RIG-I/MDA5 信号通路的中枢，其表达和功能很大程度上受到蛋白翻译后修饰的调控。进一步通过比较和分析病毒感染和表观干预对细胞基因表达的影响，研究团队发现线粒体蛋白CPT1A在病毒感染后会下调，反而在多种表观干预下均会显著上调。进一步研究发现，CPT1A是细胞静息状态下MAVS蛋白稳定表达所必需的。在表观干预激活dsRNA-RIG-I/MDA5通路时，上调的CPT1A通过提高MAVS的K63位泛素化修饰促进蛋白聚集激活，并降低K48位泛素化修饰抑制MAVS蛋白降解，进而产生以I型干扰素通路持续活化为特征的免疫激活效应，发挥了提高肿瘤微环境中CD8+ T细胞浸润和抗肿瘤免疫的作用。因此，CPT1A作为正调节因子，负责在表观干预激发抗肿瘤免疫过程中稳定MAVS的活化。在生化机制层面，线粒体定位的CPT1A通过蛋白互作的方式招募内质网定位的棕榈酰转移酶ZDHHC4来催化MAVS的79位半胱氨酸S-棕榈酰化修饰，该修饰通过影响K48位和K63位的泛素化修饰水平促进 MAVS 稳定和激活。采用CRISPRa转录激活技术提高CPT1A表达水平会进一步增加MAVS的S-棕榈酰化修饰水平，放大I型干扰素通路应答，可以使表观干预发挥更有效的抗肿瘤免疫作用。此外，CPT1A化学诱导剂，比如临床使用的非诺贝特，可以在动物模型上增强表观药物与PD-1抗体联合治疗肿瘤的效果。总之，该研究工作揭示了MAVS蛋白一种新的调控机制，其S-棕榈酰化水平显著影响表观疗法的免疫激活效应及与免疫疗法联合用药的抗肿瘤效果。图1 本研究示意图浙江大学基础医学院、医学院附属第一医院、良渚实验室生万强研究员和牛津大学Ludwig癌症研究所施扬教授为该论文的共同通讯作者，浙江大学医学院张贵恒博士研究生为该论文的第一作者。浙江大学医学院附属第四医院Dante Neculai教授，医学院附属第一医院周建娅教授和免疫所王晓健教授对本研究提供有力支持。该研究获科技部重点研发计划和基金委面上项目资助。本文仅用于学术分享，转载请注明出处。若有侵权，请联系微信：bioonSir 删除或修改！

厦门大学刘文/叶峰团队揭示促进乳腺癌进展的新机制

导读：乳腺癌（breast cancer, BC）常被称为“粉红杀手”，其发病率位居女性恶性肿瘤的首位。雌激素受体阳性乳腺癌发生和内分泌治疗抵制的分子机制尚不完全清楚。这项研究报告了circPVT1，一种由lncRNA PVT1产生的环状RNA，在ERα阳性乳腺癌细胞系和肿瘤样本中高度表达，并在促进ERα阳性的乳腺肿瘤发生和内分泌治疗抵制中起着重要作用。近日，厦门大学细胞应激生物学国家重点实验室刘文课题组与厦门大学附属第一医院叶峰课题组联合在【The EMBO Journal】杂志上发表了一篇题为“CircPVT1 promotes ER-positive breast tumorigenesis and drug resistance by targeting ESR1 and MAVS”的文章，该研究揭示了环状RNA CircPVT1在ERα阳性的乳腺癌细胞中高度表达，并且在基因转录激活和抑制中的双重功能共同促进了ERα阳性的乳腺癌的发生。https://doi.org/10.15252/embj.2022112408研究背景 01 乳腺癌雌激素受体（ER）阳性亚型约占所有乳腺癌亚型的70%，其重要的起始因素之一是雌激素的紊乱导致大量雌激素/ER靶基因的异常表达，这与细胞周期调节、细胞增殖和细胞代谢等有关。ER包括ERα和ERβ，阻断ER自身和ER通路上游或下游的治疗被称为内分泌治疗，这是ER阳性乳腺癌最重要和最独特的治疗方法。迄今为止，已经开发出了几种内分泌治疗方法。然而，由于原发性耐药，一些ER阳性乳腺癌患者无法从内分泌治疗中获益。环状RNA（circRNAs）在多种组织中高度丰富、存在并动态表达。CircRNAs在许多疾病中表现出不同的表达模式，例如癌症。近年来，越来越多的circRNAs被证明与许多生物过程和疾病高度相关。而且，一些circRNAs在ERα阳性乳腺癌细胞中高度表达。然而，这些高表达的circRNAs是否以及如何在ERα阳性乳腺癌中发挥作用尚不清楚。在这里，研究人员报告了其中一个circRNAs，即circPVT1，在ERα阳性乳腺癌细胞系和临床乳腺肿瘤样本中高度表达，这增强了ESR1系列mRNA稳定性通过海绵miR-181a-2-3p增强雌激素/ERα靶基因的激活和细胞生长。同时，circPVT1直接与MAVS相互作用，抑制RIGI和MAVS复合物的形成，抑制I型干扰素和ISG的激活以及抗肿瘤免疫。正如预期的那样，靶向circPVT1抑制ERα阳性乳腺癌细胞的生长和肿瘤发生，并克服内分泌治疗抵制。研究过程 02 该研究首先对ERα阳性乳腺癌细胞系MCF7进行了circRNA测序。并进行细胞增殖实验确定对乳腺癌细胞生长起关键作用的环状RNA。结果发现CircPVT1在乳腺癌细胞中高表达。为了进一步测试circPVT1对体内肿瘤生长的影响，使用MCF7细胞进行了异种移植实验，并使用4T1细胞进行了同种异体移植实验。结果表明，circPVT1的敲低显著减弱了肿瘤发生。CircPVT1起源于PVT1的第二个外显子，长度为410个碱基对（bps），多聚体分析结果表明，circPVT1在很大程度上与无核糖体级分相关，表明它是一种非编码RNA。数据表明，circPVT1是一种外显子circRNA，稳定地定位在细胞的细胞质中。通过敲低MCF7细胞中的circPVT1进行RNA-seq分析，发现当circPVT1在MCF7细胞衍生的异种移植物中被敲低时，代表性基因（如TFF1，PGR和GREB1）的表达受到显著抑制。综上，数据表明circPVT1激活雌激素/ ERα靶基因以促进ERα阳性乳腺肿瘤发生。为了研究circPVT1如何调节雌激素/ERα靶基因，研究人员构建竞争性内源性RNA（ceRNA）网络。网络分析表明 circPVT1 通过竞争结合 miR-181a-2-3p 稳定 ESR1 mRNA，从而促进 ERα 蛋白表达，进而促进 ERα 下游靶基因的表达。ERα 靶基因激活与细胞周期、细胞生长、代谢和耐药等密切相关。另外，circPVT1 负调控了 I 型干扰素相关基因的表达。数据还表明，circPVT1与RIGI竞争与MAVS结合，从而抑制下游信号转导和I型IFN和ISGs表达，促进了肿瘤细胞的免疫逃逸。研究意义 03 circPVT1在乳腺癌细胞和临床样本中的上调及其在ERα阳性乳腺癌中的重要作用促使研究人员研究circPVT1是否可以作为潜在的治疗靶点。根据这个思路，研究人员设计了靶向 circPVT1 的反义寡核苷酸药物（Anti-sense oligonucleotides，ASO）数据表明，ASO靶向circPVT1在抑制ERα阳性乳腺癌细胞生长和肿瘤发生方面与氟维司群一样有效，并且可以克服内分泌治疗耐药性。本研究揭示了circPVT1在基因转录激活和抑制中的双重功能共同促进ERα阳性乳腺癌的发展。ASO靶向circPVT1可有效抑制ERα阳性乳腺肿瘤生长，为乳腺癌的治疗提供了潜在的药物靶点和联合治疗策略。参考资料：https://doi.org/10.15252/embj.2022112408
