Elcatonin [60731-46-6], an analog of calcitonin, was injected i.v. into male rats daily for 63 days and into female rats daily for 14 days prior to mating.Treatment was continued in the female rats until day 7 of pregnancy. The doses were 4, 40, and 160 units/kg/day.The medium and high doses of elcatonin caused a decrease in body weight gain and a transient decrease in food intake in both male and female rats. However, elcatonin did not affect fertility or reproductive performances such as mating, impregnation, and pregnancy.Neither internal nor skeletal examination revealed any effect of elcatonin on fetal development. From these results, the maximum no-effect dose of elcatonin is estimated at 4 units/kg/day for the phys. condition of the parent rats and 160 units/kg/day for reproductive function and fetuses.
