Objective To prepare rabeprazole sodium enteric-coated capsules and inspect the formulation and quality.Methods Orthogonal experiment design L9 (34) was used to select the technol. condition of rabeprazole sodium enteric-coated pellets with the drug loading and percent yield as the indexes.Single factor experiment was used to select the formulation condition of spacer solution with the related substances as the index and select the formulation condition of enteric coating solution with the accumulative release percentage as the index.Results The technol. condition of rabeprazole sodium enteric-coated pellets were as follows: sucrose and main medicine proportion was 6:1, the coating granulator speed was 100 r·min-1, and microcrystalline cellulose and the main medicine proportion was 3:1.The 12% HPMC was used as the spacer material and 15% Eudragit L30D-55 as the enteric coating material.The 3 batches of prepared rabeprazole sodium enteric-coated capsules had good stability, with resistance to acid all higher than 95% and 30 min dissolution rate all higher than 98%.The content and related substances all met the requirements.The accelerate test of acid tolerance rate, the dissolution rate, the content and related substances all met the requirements.Conclusion Verified by 3 batches of products and accelerated tests, the products had good enteric and drug release properties with good stability.
