To study the optimum purified technol. condition of flavonoid extract from Platycladus orientalis leaves, the suitable type of macroporous resin was selected through static experiment Then, the influence of sample solution pH value, sample concentration, loading velocity, sample volume, volume fraction of eluent, eluent volume and elution velocity on performance of adsorption and desorption of flavonoid extract from Platycladus orientalis leaves were investigated resp. by the dynamic experiment The results showed that the AB-8 macroporous resin was suited for flavonoid extract purification from Platycladus orientalis and optimum purification process condition was established as follows: 3 mg·mL-1 loading sample concentration that solution pH value of 6.0 was loaded to the column at velocity of 2.0 mL·min-1 in volume of 60 mL, then 70% (V/V) alc. that volume of 140 mL was applied to elution with the velocity of 1.0 mL·min-1. After purification, the purity of flavonoid in sample increased from (24.7± 0.8)% to (68.2±1.1)%, therefore this technol. condition had better purification efficiency, could be used for enrichment of flavonoids of Platycladus orientalis leaves.
