Ubiquitin-like modifier involved in formation of autophagosomal vacuoles (autophagosomes) (PubMed:20418806, PubMed:23209295, PubMed:28017329). Plays a role in mitophagy which contributes to regulate mitochondrial quantity and quality by eliminating the mitochondria to a basal level to fulfill cellular energy requirements and preventing excess ROS production (PubMed:23209295, PubMed:28017329). In response to cellular stress and upon mitochondria fission, binds C-18 ceramides and anchors autophagolysosomes to outer mitochondrial membranes to eliminate damaged mitochondria (PubMed:22922758). While LC3s are involved in elongation of the phagophore membrane, the GABARAP/GATE-16 subfamily is essential for a later stage in autophagosome maturation (PubMed:20418806, PubMed:23209295, PubMed:28017329). Promotes primary ciliogenesis by removing OFD1 from centriolar satellites via the autophagic pathway (PubMed:24089205). Through its interaction with the reticulophagy receptor TEX264, participates in the remodeling of subdomains of the endoplasmic reticulum into autophagosomes upon nutrient stress, which then fuse with lysosomes for endoplasmic reticulum turnover (PubMed:31006537, PubMed:31006538). Upon nutrient stress, directly recruits cofactor JMY to the phagophore membrane surfaces and promotes JMY's actin nucleation activity and autophagosome biogenesis during autophagy (PubMed:30420355).

Novel insights into copper-induced Chinese mitten crab hepatopancreas mitochondrial toxicity: Oxidative stress, apoptosis and BNIP3L-mediated mitophagy

Copper (Cu) is an important metal pollutant commonly found in aquatic environment. Cu-based nanoparticles (NPs) have been increasingly fabricated, and led to cytotoxicity in aquatic animals. Herein, the mechanisms underlying the CuSO₄/Cu-NPs-mediated perturbation of the hepatopancreatic mitochondrial function at different concentrations were investigated and compared. After exposing Eriocheir sinensis to 0 (control), 5, 50, and 500 μg/L CuSO₄ and 10 μg/L Cu-NPs for 21 days, hepatopancreases were retrieved. The results revealed that Cu-NPs or excess CuSO₄ induced ultrastructural damage following a time-dose effect, as indicated by swelling and degeneration of the lumen of hepatic tubules. Cu-NPs or excess CuSO₄ exposure decreased the antioxidative capacity and led to the over-accumulation of reactive oxygen species (ROS). Moreover, the mitochondrial membrane potential (MMP) was reduced and apoptosis induced. Additionally, both CuSO₄ and Cu-NPs increased the numbers of mitophagosomes and the mRNA and protein levels of microtubule associated protein 1 light chain 3 beta (LC3B), and triggered mitophagy through BCL2 interacting protein 3 like (BNIP3L)/Beclin1 pathway. Altogether, this study provides a basis for exploring Cu-mediated potential mitochondrial autophagy activation mechanisms, uncovered the difference between CuSO₄ and Cu-NPs.

Lysosomal damage due to cholesterol accumulation triggers immunogenic cell death

Cholesterol serves as a vital lipid that regulates numerous physiological processes. Nonetheless, its role in regulating cell death processes remains incompletely understood. In this study, we investigated the role of cholesterol trafficking in immunogenic cell death. Through cell-based drug screening, we identified two antidepressants, sertraline and indatraline, as potent inducers of the nuclear translocation of TFEB (transcription factor EB). Activation of TFEB was mediated through the autophagy-independent lipidation of MAP1LC3/LC3 (microtubule associated protein 1 light chain 3). Both compounds promoted cholesterol accumulation within lysosomes, resulting in lysosomal membrane permeabilization, disruption of autophagy and cell death that could be reversed by cholesterol depletion. Molecular docking analysis indicated that sertraline and indatraline have the potential to inhibit cholesterol binding to the lysosomal cholesterol transporters, NPC1 (NPC intracellular cholesterol transporter 1) and NPC2. This inhibitory effect might be further enhanced by the upregulation of NPC1 and NPC2 expression by TFEB. Both antidepressants also upregulated PLA2G15 (phospholipase A2 group XV), an enzyme that elevates lysosomal cholesterol. In cancer cells, sertraline and indatraline elicited immunogenic cell death, converting dying cells into prophylactic vaccines that were able to confer protection against tumor growth in mice. In a therapeutic setting, a single dose of each compound was sufficient to significantly reduce the outgrowth of established tumors in a T-cell-dependent manner. These results identify sertraline and indatraline as immunostimulatory agents for cancer treatment. Human antigen R -mediated autophagy-related gene 3 methylation enhances autophagy-driven ferroptosis in Crohn's disease colitis

BACKGROUND AND AIMSCrohn's disease (CD) is a chronic inflammatory disorder that can affect any part of the gastrointestinal tract, with the exact etiology remaining unclear. Recent studies have implicated the role of human antigen R (HuR) in the pathogenesis of various inflammatory diseases, including CD. However, the role of HuR in the modulation of CD remains underexplored. Therefore, this study aimed to investigate the mechanistic involvement of HuR in CD.METHODSWe established colitis models using human intestinal epithelial cells and lipopolysaccharide and dextran sulfate sodium-induced mice. Additionally, by knocking out HuR in both cell and animal models, we validated the role of HuR in autophagy and ferroptosis. The role of HuR in regulating ferroptosis accompanied by autophagy activation in CD was detected using ELISA, flow cytometry, immunofluorescence, transmission electron microscopy, Western blot, and RT-qPCR. The demethylation level of ATG3 and the stability of ATG3 mRNA regulated by HuR were detected using immunofluorescence, RIP, and MeRIP-qPCR. The effect of HuR on DSS-induced colitis was evaluated using DAI score, H&E staining, TUNEL staining, and immunohistochemistry.RESULTSThe results show that HuR expression is significantly increased in CD colonic inflammation. Compared with the control group, the model group mice exhibited decreased levels of lipid peroxidation markers glutathione and superoxide dismutase, elevated malondialdehyde and reactive oxygen species levels, and reduced expression of iron-related proteins glutathione peroxidase 4, ferritin heavy chain protein 1, and solute carrier family 7 member 11. Additionally, the expression of autophagy-related proteins microtubule-associated protein 1 A/1B-light chain 3, beclin-1, and autophagy related 3 (ATG3) was upregulated, while p62 expression was downregulated. In both in vitro and in vivo models, HuR knockout reversed these changes induced by lipopolysaccharide and dextran sulfate sodium, concomitant with improved tissue pathology. Mechanistically, HuR enhances autophagy-mediated ferroptosis in CD colonic inflammation by regulating ATG3 methylation and mRNA stability.CONCLUSIONHuR accelerates colonic inflammation in CD by regulating ATG3 methylation, which enhances autophagy-mediated ferroptosis. Knockout of HuR alleviates Crohn's colitis. This finding provides a potential therapeutic target for the treatment of CD.

Acta Pharmacologica Sinica: 氯喹通过抑制自噬途径提高化疗药物治疗急性髓系白血病的疗效

急性髓系白血病(AML)是一种骨髓和外周血中髓系祖细胞的异质性恶性肿瘤，以克隆性髓系母细胞快速聚集、分化不成熟为特征。随着对其分子生物学和遗传学基础认识的增加，新的治疗方法取得了巨大的进步。 AML的一般治疗选择是诱导治疗、巩固治疗和造血细胞移植。这些治疗方案中使用的大多数药物是阿糖胞苷、柔红霉素和伊达比星。然而，AML的完全缓解(CR)率不容乐观。青壮年一线治疗CR率为60%~80%，65岁以上老年人CR率为40%~60%。同时，采用当前治疗策略治疗的急性髓细胞白血病患者往往缓解时间较短(少于6个月)，并且大多数在3年内复发。近日，来自复旦大学的研究者们在Acta Pharmacologica Sinica杂志上发表了题为“Chloroquine enhances the efficacy of chemotherapy drugs against acute myeloid leukemia by inactivating the autophagy pathway”的文章，该研究揭示了氯喹通过抑制自噬途径提高化疗药物治疗急性髓系白血病的疗效。目前急性髓系白血病(AML)的治疗在很大程度上受到阿糖胞苷、柔红霉素和伊达比星等常用化疗药物耐药性的阻碍。在这项研究中，研究者研究了化疗耐药的分子机制，以及提高这些药物对AML疗效的潜在策略。研究者通过分析AML的体外药物反应和多组学分析公开数据，我们确定自噬激活是化疗耐药患者的一个潜在靶点。在THP-1和MV-4-11细胞系中，自噬调节基因ATG5或MAP1LC3B的敲除显著增强了AML细胞对化疗药物阿糖胞苷、柔红霉素和伊达比星的敏感性。在计算机筛选中，研究者发现磷酸氯喹模拟自噬失活。研究者发现磷酸氯喹呈剂量依赖性地下调MV-4-11细胞的自噬途径。此外，磷酸氯喹与化疗药物在体内外均有协同抗肿瘤作用。这些结果表明自噬激活是一种耐药机制，磷酸氯喹和化疗药物联合治疗可以增强抗AML的效果。综上所述，本研究确定自噬激活是AML对一线化疗药物耐药的关键机制，并进一步确定了现有的针对自噬途径的药物氯喹与化疗药物联合使用可以克服耐药性，提高抗AML的疗效。本研究建议未来的临床研究评估化疗药物的联合治疗和抑制自噬作为下一代AML的治疗策略。（生物谷 Bioon.com）参考文献 Han-lin Wang et al.
