Objective To establish a ultra performance liquid chromatog.-evaporative light scattering detector (UPLC-ELSD) method for determining the ginsenoside Rg1 and Re contents in Shensong Yangxin Capsule.Methods The Waters ACQUITY UPLC BEH C18 column (100 mm × 2.1 mm, 1.7 μm) was adopted.The mobile phase consisted of acetonitrile-water with gradient elution.The flow rate was 0.45 mL/min, the column temperature was 35°C and the injection volume was 2 μL.As to the ELSD, the temperature of drift tube was 50°C and the flow rate of carrier gas was 30.0 psi.Results The sample seizes of ginsenoside Rg1 and Re in Shensong Yangxin Capsule within the range of 0.09108-1.1358 μg and 0.08306-1.03825 μg had good linear relation with the peak area, resp.And the average recovery rates were 99.01% and 99.25%, RSD 0.50% and 0.37% (n = 6), resp.Conclusion Compared with HPLC, the UPLC method may greatly improve the anal. speed of ginsenoside Rg1 and Re in Shensong Yangxin Capsule without affecting the separation effect, at the same time reduce the solvent consumption.As an alternative of conventional HPLC, UPLC is more convenient, rapid and reliable.
