An HPLC method for the assay of glutathione (GSH) and determination of glutathione oxidized (GSSG) in reduced glutathione in injection was presented.A Waters Nova-Pak C18 column (150 mm x 3.9 mm, 4 μm) was used, the mobile phase was 0.01 mol·L-1 sodium heptanesulfonate in 0.05 mol·L-1 potassium dihydrogen phosphate solution (pH 3.0)-methanol (97:3, V/V), the detection wavelength was 210 nm, the column temperature was 30°C.The calibration curve was linear within the range of 0.01 - 1 g·L-1 for GSH, with regression equation being A = 9.535 6 × 106c + 529 62 (r = 0.999 9, n = 6), the average recovery 99.72% (RSD = 0.43%, n = 9).The intra-day and the inter-day precisions were 0.41% (n = 6) and 0.49% (n = 5), resp.Determination limit of GSSG was 1 ng (rSN = 3), quantitation limit was 5 ng (rSN = 10), the average recovery was 99.0% (RSD = 0.61%, n = 9), the calibration curve was linear within the range of 0.5 - 50 mg·L-1 for GSSG, with regression equation being A = 10 054c + 296 (r = 1.000 0, n = 7).This method is simple, specific, accurate, reliable and can be used in the quality standard of the drug.
