A test based on the estimation of urinary pregnanediol excretion is described, which can be carried out in 2-3 hrs.A 100 ml. urine sample acidified with HCl is extracted with cyclohexane.The concentrated extract is applied to the silica-gel sheets described by Stahl (CA 53, 27c), which are developed for 20 min. with a 9:1 mixture of CHCl3 and Me2CO.The plates are sprayed with approx. 40% H3PO4 and, after drying for 5 min. at 100-120°, with a 1.5% alc. solution of phosphomolybdic acid.By this technique, pregnanediol can be detected in urine from the 2nd to 3rd month of pregnancy.
