To prevent and control gout in geese caused by Goose astrovirus (GAstV), preparation of inactivated vaccine, immunization of hens and preparation of yolk antibody by acidified water-caprylic acid method were performed in this study. Neutralization titer and purity of yolk antibody prepared with chloroform, polyethylene glycol and ammonium sulfate were compared in neutralization test and SDS-PAGE test, then the safety and efficacy of the yolk antibody were evaluated. The results of different immunization procedures for hen showed that inactivated vaccines immunized three times, immunization interval 21 days and the immunization doses of 1.0 mL, 1.5 mL and 2.0 mL, resp. was the best immune procedure. The neutralization test results showed that the neutralization titer of yolk antibody were 1:445.7 prepared by acidified water-caprylic acid method, and there were few heteroproteins in protein electrophoresis except the target band in the protein electrophoresis. The titer and purity of yolk anti-body were higher than other three methods. The safety tests showed that when goslings were inoculated with single dose (0.5 mL), all the goslings were alive and no adverse reaction was observed All the results indicated a good safety of the yolk antibody. The potency results showed that no less than 80% goslings were protected, when the goslings were inoculated with yolk antibody of titer of 1:128 attacked with GAstV (1000 ELD50) after 24 h of inoculation. Moreover, the min. immune dose was 0.25 mL per gosling of 3-day-old age and 0.5 mL per gosling of 4-day-old age and 7-day-old age. In generation and immune duration tests, goslings could obtain effective passive protection (protection rate ≥80%), when they were attacked with GAstV (1000 ELD50) after eight hours to five days inoculated with yolk antibody. Namely, the immune production period was eight hours after antibody inoculation, and the immune duration persisted 5 days. The yolk antibody prepared in this study was safe and effective, and could provide effective prevention and treatment means for the prevention and control of goose gout disease in China.
